Favorite JavaScript framework?

I'm getting ready to start developing a new web application, from the ground up. It's going to be heavily interactive, full of AJAX and all that other "Web 2.0" stuff, yadda yadda yadda.

Anyway, the point is there'll be much client-side scripting going on, so I'm curious what people use and/or recommend for a JavaScript framework that (at a bare minimum) supports DOM scripting and AJAX.

I have some experience using Prototype, but I'm not married to it by any stretch of the imagination and am curious what thoughts/experiences others have.
